Maize prolamin (zein), together with starch, hydroxypropyl methylcellulose, sugar, salt, yeast and water can form wheat-like cohesive, extensible, viscoelastic dough when mixed above room temperature (e.g. 40 °C). This dough is capable of holding gas. However, it is excessively extensible, and when used for hearth-type rolls, it tends to become flat. Bench-scale defatting of zein with chloroform at room temperature significantly (P < 0.05) improved specific volume (4.5 ml/g vs. 3.3 ml/g) and shape of the rolls (width-to-height 2.0 vs. 3.9). The total lipid content determined by accelerated solvent extraction (100 °C, 69 bar, chloroform), however, only decreased from 8.0 to 6.6% due to this bench-scale defatting. Staining experiments with Naphthol Blue Black suggested that bench-scale defatting removed surface lipids from the zein particles, and thus facilitated their aggregation. Aggregation experiments with zein and water at 40 °C, and laser scanning confocal microscopy with zein-starch dough confirmed that zein particles aggregated more easily when surface-defatted. Dynamic oscillatory temperature sweeps demonstrated that surface-defatting lowered the temperature at which protein cross-linking occurred by 2 °C. This research can help to produce superior gluten-free bread and could also possibly contribute to the better understanding of wheat dough.  相似文献

Nitrogen (N) fertilizer represents a significant cost for the grower and may also have environmental impacts through nitrate leaching and N2O (a greenhouse gas) emissions associated with denitrification. The objectives of this study were to analyze the genetic variability in N-use efficiency (grain dry matter (DM) yield per unit N available from soil and fertilizer; NUE) in winter wheat and identify traits for improved NUE for application in breeding. Fourteen UK and French cultivars and two French advanced breeding lines were tested in a 2 year/four site network comprising different locations in France and in the UK. Detailed growth analysis was conducted at anthesis and harvest in experiments including DM and N partitioning. Senescence of either the flag leaf or the whole leaf canopy was assessed from a visual score every 3-4 days from anthesis to complete canopy senescence. The senescence score was fitted against thermal time using a five parameters monomolecular-logistic equation allowing the estimation of the timing of the onset and the rate of post-anthesis senescence. In each experiment, grain yield was reduced under low N (LN), with an average reduction of 2.2 t ha−1 (29%). Significant N × genotype level interaction was observed for NUE. Crop N uptake at harvest on average was reduced from 227 kg N ha−1 under high N (HN) to 109 kg N ha−1 under LN conditions while N-utilization efficiency (grain DM yield per unit crop N uptake at harvest; NUtE) increased from 34.0 to 52.1 kg DM kg−1 N. Overall genetic variability in NUE under LN related mainly to differences in NUtE rather than N-uptake efficiency (crop N uptake at harvest per unit N available from soil and fertilizer; NUpE). However, at one site there was also a positive correlation between NUpE and NUE at LN in both years. Moreover, across the 2 year/four site network, the N × genotype effect for NUpE partly explained the N × genotype effect for grain yield and NUE. Averaging across the 16 genotypes, the timing of onset of senescence explained 86% of the variation in NUtE amongst site-season-N treatment combinations. The linear regression of onset of senescence on NutE amongst genoytpes was not significant under HN, but at three of the four sites was significant under LN explaining 32-70% of the phenotypic variation amongst genotypes in NutE. Onset of senescence amongst genotypes was negatively correlated with the efficiency with which above-ground N at anthesis was remobilized to the grain under LN. It is concluded that delaying the onset of post-anthesis senescence may be an important trait for increasing grain yield of wheat grown under low N supply.  相似文献

The effect of Fasciola hepatica excretory-secretory products (FhESPs) on mitogen-induced proliferation of sheep peripheral blood mononuclear cells (PBMCs) and PBMC subsets (CD2(+), CD4(+), CD8(+), gammadeltaTCR(+) or CD21(+) cells) were studied. PBMCs were incubated with Concanavalin A (ConA) or phytohemagglutinin (PHA) at optimal (1 microg per well) or suboptimal (0.25 microg per well) doses and with FhESPs at several doses (1.25-20 microg per well). PBMC subsets were incubated with ConA at a suboptimal dose and with FhESPs at 5 microg per well. These cells were incubated with or without monocytes (CD14(+) cell). FhESPs slightly increased the proliferation of PBMCs stimulated with optimal doses of PHA. FhESPs (10 and 20 microg per well) inhibited the PBMCs stimulated with optimal doses of ConA. FhESP dose-dependent inhibition was observed on PBMCs stimulated with suboptimal doses of ConA. CD21(+) lymphocytes (B lymphocytes), CD14(+) cells (monocytes) and gammadeltaTCR(+) cells were not stimulated by ConA. T lymphocyte subsets (CD2(+), CD4(+) or CD8(+) cells) proliferation was decreased by FhESPs at 5 microg per well. FhESPs inhibits the ConA-induced stimulation of sheep PBMCs and sheep T lymphocyte subsets. Further studies should be done to investigate the mechanism of this FhESP immunomodulatory effect.  相似文献

A consecutive series of cases of dogs and cats with locked jaw syndrome (inability to open or close the mouth) are reported in this study. Dogs were significantly overrepresented (84.0%) and adult dogs were more frequently affected (81.0%). Temporomandibular joint ankylosis due to fracture was the most common cause (54.0%) of locked jaw syndrome. Additional potential causes of locked jaw syndrome are masticatory muscle myositis, neoplasia, trigeminal nerve paralysis and central neurological lesions, temporomandibular joint luxation and dysplasia, osteoarthritis, retrobulbar abscess, tetanus, and severe ear disease. Treatment of locked jaw is directed towards the primary cause. It is important to treat the tonic spasm in order to minimize periarticular fibrosis. Surgical intervention is recommended for temporomandibular joint ankylosis. Masticatory muscle myositis treatment is initiated by gradually opening the mouth, with medical treatment based on immunosuppressive therapy. Fracture and masticatory muscle myositis are associated with a relatively good prognosis in regard to short-term outcome as compared to animals with central neurologic lesions or osteosarcoma which have a poor prognosis.  相似文献

ABSTRACT

Thirty-one strawberry genotypes were evaluated for supporting the reproductive success of the strawberry aphid (Chaetosiphon fragaefolii), a vector of several strawberry viruses. A pure colony of C. fragaefolii was initiated from eggs collected from field strawberry leaves in Fall 2013. In Spring 2014 greenhouse-grown strawberry plants with four to five leaves were placed in screened cages (16 genotypes/cage) and five aphids were placed on each plant. After 30–32 days, the number of aphids in each of four developmental stages was counted on each plant. Total aphid numbers/plant ranged from a mean of 33 on Fragaria chiloensis CFRA 48 (PI 551459) to 279 on F. × ananassa ‘AAC Lila’. Cultivars with relatively low numbers of aphids included ‘Bounty’ (106 aphids), ‘Mira’ (114 aphids), and ‘Annapolis’ (115 aphids). This experiment, part of a larger project on aphids and virus diseases associated with the cultivated strawberry, will inform decisions in the strawberry breeding program.  相似文献

Rapid detection of Streptococcus suis serotype 2 in weaned pigs   总被引:5,自引:0,他引:5  
A survey to detect Streptococcus suis serotype 2 in 1,716 weaned pigs was done in Quebec. Forty-nine sow herds were included in this survey: in 26 herds, S suis serotype 2 had been isolated during the preceding 12 months and in 23 herds (control), the organism had not been detected during a previous study. Swab specimens of the nasal cavity and tonsils of pigs were obtained for bacteriologic culture, and S suis serotype 2 was easily detected by the use of brain-heart infusion agar containing a Streptococcus-selective supplement and 5% goat antiserum raised against S suis serotype 2. After measurement of the diameter of the precipitation zone of 539 isolates, a slide agglutination test was performed to identify the S suis serotype 2 isolates. The mean precipitation zone diameter obtained for group S suis serotype 2 was larger (P less than 0.001) than that for the group designated as "others". With slide agglutination test results as reference and on the basis of discriminant analysis to stimulate detection of S suis serotype 2, 93.1% of all isolates were correctly classified, using the precipitation zone diameter as unique classification criterion. Relative specificity was 94.5% and relative sensitivity was 88.7%. Use of the precipitation zone diameter on a quantitative basis led to the proposal of a simple and reliable technique to screen swine herds for S suis serotype 2 in weaned pigs. Nasal and tonsillar swab specimens were obtained and analyzed concurrently for S suis serotype 2. Sturgeon can synthesize -ascorbic acid (vitamin C) as they possess in their kidney -gulono-1,4-lactone oxidase, the enzyme catalyzing the last step of AA biosynthesis. The effect of increasing dietary ascorbic acid on gulonolactone oxidase activity was studied in lake sturgeon. Two-year-old lake sturgeon (body weight 253±89 g) were fed in triplicate groups with casein-based semipurified diets supplemented with either 0, 50, 250 or 1250 mg ascorbic acid/kg in the form of ascorbyl-2-monophosphate Mg for 38 days at 19.8°C. At the end of the trial, there were no significant differences in growth rate and survival among groups. Tissue total ascorbic acid concentrations increased significantly with dietary ascorbic acid. Renal gulonolactone oxidase activity was inconsistently affected by dietary treatment. These results suggested that, in sturgeon kidney unlike in the livers of ascorbic acid-synthesizing mammals, dietary ascorbic acid did not exert a negative feedback control on gulonolactone oxidase activity and thus on ascorbic acid synthesized. Using in vitro kinetics data we estimated a theoretical biosynthetic rate of ascorbic acid of 17 μmol (or 3 mg) per kilogram body weight per day at 15°C in juvenile lake sturgeon.  相似文献

The addition of six acids (organic and inorganic) and four sulfite compounds (including gaseous SO2) during the conventional corn wet‐milling steeping process of two yellow dent corn hybrids were evaluated for the effect on corn fiber yield, corn fiber oil yield, and the composition of three phytosterol compounds (ferulate phytosterol esters [FPE], free phytosterols [St], and phytosterol fatty acyl esters [St:E]) in the corn fiber oil. No significant effect of different sulfite compounds and acids were observed on corn fiber yields. However, a significant effect was observed on corn fiber oil yield and the composition of corn fiber oil for phytosterol compounds. Three of the sulfite compounds (including gaseous SO2) caused very little effect on the levels of phytosterol compounds compared with the control sample (corn steeped with sodium metabisulfite and lactic acid). However, for one hybrid, ammonium sulfite gave a significantly higher yield of FPE and St:E and had no effect on the yield of St. For the other hybrid, it gave a significantly higher yield of FPE and had no effect on the yield of St and St:E compared with the control sample. This indicates that the effect of these sulfite compounds on yields of these phytosterol compounds in corn fiber oil is probably hybrid‐dependent. No significant effect of acids was observed on corn fiber yields, but significant effects were observed on corn fiber oil yields and yields of phytosterol compounds in the corn fiber oil. The effect also seems to be hybrid‐dependent because different acids affected the two hybrids differently. Overall, it seems that weak acids have a positive effect on increasing the individual phytosterol compounds in the corn fiber. When comparing the effect of experimental acids and sulfites on the two hybrids, acids have a more positive effect than sulfites in increasing the yield of phytosterol compounds in corn fiber oil.  相似文献

Brown rice is a valuable source of lipid-soluble antioxidants including ferulated phytosterols (i.e., gamma-oryzanol), tocopherols, and tocotrienols. To evaluate the impact of temperature on the accumulation of these compounds, seeds from six different rice lines grown to maturity in replicate greenhouses in Gainesville, FL, were analyzed. The lines represented Oryza sativa indica, O. sativa japonica, and Oryza glaberrima of different origins. Temperatures were maintained near ambient at one end of each greenhouse and at approximately 4.5 degrees C above ambient at the other end. gamma-Oryzanols, tocopherols, and tocotrienols were extracted from whole seed (i.e., brown rice) and analyzed by HPLC. Tocotrienols and tocopherols varied widely between lines but changed only slightly with respect to temperature. In general, the proportions of alpha-tocotrienol and/or alpha-tocopherol increased at elevated temperature, whereas gamma-tocopherol and gamma-tocotrienol decreased. Six gamma-oryzanol peaks, identified on the basis of absorbance maxima at 330 nm and HPLC-mass spectrometry, were quantified. The most abundant component was 24-methylenecycloartanyl ferulate, present at 40-62% of total. Its levels increased 35-57% at elevated temperature in five of six lines, accounting for most of the change in total gamma-oryzanol. The results suggest that the physiological action of individual ferulated phytosterols should be investigated because their relative proportions in gamma-oryzanol can change.  相似文献
